For a first-timer, Panamá is going to be the best of the three. Best infrastructure and broadband access. Relatively safe compared to the other two on your list. Punta Pacifica and Punta Paitilla are nice areas, though not the cheapest. Hostal Casa Areka is a good place to stay if you're on a budget. Nicaragua is alright, I spent some time there on a consulting project. Managua is not the most interesting city, but Granada and San Juan del Sur are nicer, but internet and infrastructure are not as good as Panamá. I haven't been to Ecuador, but I'd recommend building up some more LatAm experience before considering it.

If you go with the Ecuador option, Quito + Cuenca is a very smooth combo for a short 3–5 day digital nomad trip. A couple of quick practical thoughts on Ecuador for remote work: In Cuenca (especially in El Centro / Historic Center), fiber internet is standard and very fast. I get 100+ Mbps up/down where I live. Most modern apartments and local cafes handle video calls with zero issues. There is a solo Nomad Vibe. I meet a ton of solo travelers. It’s extremely walkable, very safe, and has a great cafe culture (Café San Sebas, Café Ñucallacta, etc.) where tons of solo remote workers hang out during the day. The quick 45-minute flight between Quito and Cuenca is pretty nice. September weather here is generally pleasant for working with a mountain view in the morning and exploring historic streets or local spots in the afternoon.

Nicaragua WiFi is truly hit-or-miss except at hostels; Panama City has its coworking sorted out. Ecuador was a winner for me; Banos had good WiFi and the Quito-Banos circuit took care of five days nicely. San Juan, if you ever go there, Las Ruinas does the hot-cold plunge I was planning for my recovery day

Then I'd live on just over 15k and invest the rest, but that's just me. It'd still leave 1.278k/month, more than 1.7k USD. Definitely more limiting, I'd forget pretty much all of Europe, especially since you mentioned not wanting smaller towns. I'd go to Bolivia and chill and stay as long as possible. I wouldn't really have to worry about money since I'd have 2x the budget required, would amortize the flights expense over several months and build up some cash reserves (separate from the initial 1/3 savings which aren't to be touched). When the time came to move on, I'd do so by land to keep travel costs down. Brazil if looking for more of a change (not Rio/São Paulo), Ecuador (not Quito) if wanting to stay in a Spanish speaking country.

Copa will let you stop in Panama without penalty on your way to another Latin American city. Get a trip to a cheaper destination; it will stop in Panama, yes or yes, so you do the free stopover Over for a few days and you can save quite a bit more in airfare. But I would skip Panama in September-October; it rains quite a bit, usually for days, with no stop. I am not the biggest fan of Panama City, but the rest of the country is worth every stupidly expensive dollar you would have spent in the city, with lots of adventures and things to see and do. September is the height of the storm season in the Dominican Republic; also skip it unless you are going to be staying for more than a few days. I would choose Ecuador, but try to go to Guayaquil instead of Quito if I were you. Volcano boarding is a must-do in Nicaragua.
